Uncertain significance for Deficiency of UDPglucose-hexose-1-phosphate uridylyltransferase — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000155.4(GALT):c.844C>G (p.Leu282Val), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces leucine with valine at codon 282 of the GALT protein (p.Leu282Val). The leucine residue is highly conserved and there is a small physicochemical difference between leucine and valine. This variant is not present in population databases (ExAC no frequency). This missense change has been observed in individual(s) with galactosemia (PMID: 11261429; Invitae). In at least one individual the data is consistent with the variant being in trans (on the opposite chromosome) from a pathogenic variant. ClinVar contains an entry for this variant (Variation ID: 25276). Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Possibly Damaging"; Align-GVGD: "Class C0"). Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
