Uncertain significance — the classification assigned by Ambry Genetics to NM_006648.4(WNK2):c.531G>C (p.Glu177Asp), citing Ambry Variant Classification Scheme 2023: The p.E177D variant (also known as c.531G>C), located in coding exon 1 of the WNK2 gene, results from a G to C substitution at nucleotide position 531. The glutamic acid at codon 177 is replaced by aspartic acid, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
