This variant lies in the LDLR gene (transcript NM_000527.5) at coding-DNA position 1754, where T is replaced by C; at the protein level this means replaces isoleucine at residue 585 with threonine — a missense variant. Submitter rationale: Variant summary: LDLR c.1754T>C (p.Ile585Thr) results in a non-conservative amino acid change in the encoded protein sequence. Algorithms developed to predict the effect of missense changes on protein structure and function all suggest that this variant is likely to be disruptive. The variant was absent in 251492 control chromosomes (gnomAD). c.1754T>C has been observed in multiple individuals affected with Familial Hypercholesterolemia and this variant co-segregated with the disease (Widhalm_2007, Maglio_2014, Marusic_2020, Klanar_2015). These data indicate that the variant is very likely to be associated with disease. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
