NM_000527.5(LDLR):c.947A>C (p.Asn316Thr) was classified as Uncertain Significance for Hypercholesterolemia, familial, 1 by All of Us Research Program, National Institutes of Health, citing ACMG Guidelines, 2015: This missense variant replaces asparagine with threonine at codon 316 of the LDLR protein. This variant is also known as p.Asn295Thr in the mature protein. Computational prediction suggests that this variant may have a deleterious impact on protein structure and function (internally defined REVEL score threshold >= 0.7, PMID: 27666373). To our knowledge, functional studies have not been reported for this variant. This variant has been reported in several individuals affected with familial hypercholesterolemia (PMID: 21310417, 22698793; ClinVar SCV000540775.1). This variant has not been identified in the general population by the Genome Aggregation Database (gnomAD). A different variant affecting the same codon, p.Asn316Ser, is considered to be disease-causing (ClinVar variation ID: 183103), suggesting that asparagine at this position is important for LDLR protein function. The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.
