Uncertain significance — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_000552.5(VWF):c.236G>T (p.Gly79Val), citing LabCorp Variant Classification Summary - May 2015: Variant summary: VWF c.236G>T (p.Gly79Val) results in a non-conservative amino acid change located in the von Willebrand factor, type D domain (IPR001846) of the encoded protein sequence. Four of five in-silico tools predict a damaging effect of the variant on protein function. The variant was absent in 251460 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.236G>T has been reported in the literature in at least one individual with unknown zygosity affected with Von Willebrand Disease (Veyradier_2016). These report(s) do not provide unequivocal conclusions about association of the variant with Von Willebrand Disease. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
