Uncertain significance for Cardiomyopathy — the classification assigned by Color Diagnostics, LLC DBA Color Health to NM_001035.3(RYR2):c.10936-2A>G, citing ACMG Guidelines, 2015. This variant lies in the RYR2 gene (transcript NM_001035.3) at the canonical splice acceptor site of the intron immediately before coding-DNA position 10936, where A is replaced by G; at the protein level this means a change at this position may disrupt normal splicing. Submitter rationale: This variant causes an A to G nucleotide substitution at the -2 position of intron 77 of the RYR2 gene. Splice site prediction tools predict that this variant may have a significant impact on RNA splicing. To our knowledge, RNA studies have not been reported for this variant. This variant has not been reported in individuals affected with RYR2-related disorders in the literature. This variant has not been identified in the general population by the Genome Aggregation Database (gnomAD). The role of loss-of-function RYR2 truncation and splice variants in autosomal dominant cardiovascular disorders is not clearly established. The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.
