Uncertain significance for Hereditary spherocytosis type 1 — the classification assigned by Diagnostics Services (NGS), CSIR - Centre For Cellular And Molecular Biology to NM_000037.4(ANK1):c.3598T>C (p.Cys1200Arg), citing ACMG Guidelines, 2015. This variant lies in the ANK1 gene (transcript NM_000037.4) at coding-DNA position 3598, where T is replaced by C; at the protein level this means replaces cysteine at residue 1200 with arginine — a missense variant. Submitter rationale: The c.3598T>C variant is not present in publicly available population databases like 1000 Genomes, ExAC, EVS, gnomAD, Indian Exome Database or our in-house exome database. This variant has neither been published in literature nor reported to clinical databases like in ClinVar, Human Gene Mutation Database (HGMD) or OMIM, in any affected individuals. In silico pathogenicity prediction programs like SIFT, PolyPhen2, MutationTaster2, CADD, etc predicted this variant to be likely deleterious, however these predictions were not confirmed by published functional studies.
