Uncertain significance — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NC_000017.10:g.(41199721_41201137)_(41277501_?)dup, citing LabCorp Variant Classification Summary - May 2015: Variant summary: The variant identified by MLPA or other technology involves the duplication of exons 1-21 in the BRCA1 gene, where exon 2 contains the initiation codon. A presumed nomenclature of c.(?_-233)_(5406+1_5407-1)dup has been designated for the purposes of this classification. It has been assumed that this is a tandem duplication in direct orientation (Richardson_GIM_2018, Newman_AJHG_2015). The variant was absent in 21694 control chromosomes (gnomAD, Structural Variants dataset). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.(?_-233)_(5406+1_5407-1)dup has been reported in the literature in at least one individual affected with Hereditary Breast And Ovarian Cancer Syndrome, however without strong evidence for causality (Rebbeck_2018). This report does not provide unequivocal conclusions about association of the variant with Hereditary Breast And Ovarian Cancer Syndrome. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. The following publication has been ascertained in the context of this evaluation (PMID: 29446198). Two ClinVar submitters (evaluation after 2014) have cited the variant: one submitter classified the variant as pathogenic, and one submitter classified the variant as uncertain significance. Based on the evidence outlined above, the variant was classified as uncertain significance.
