Uncertain significance — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_001206744.2(TPO):c.1970T>C (p.Ile657Thr), citing LabCorp Variant Classification Summary - May 2015: Variant summary: TPO c.1970T>C (p.Ile657Thr) results in a non-conservative amino acid change in the encoded protein sequence. Five of five in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 2.4e-05 in 251290 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.1970T>C has been reported in the literature in at least two compound heterozygous individuals affected with congenital hypothyroidism (e.g., Ma_2015, Narumi_2017 (meeting abstract)). These data indicate that the variant may be associated with disease. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. The following publication was ascertained in the context of this evaluation (PMID: 26174974). No clinical diagnostic laboratories have submitted clinical-significance assessments for this variant to ClinVar after 2014. Based on the evidence outlined above, the variant was classified as VUS-possibly pathogenic.
