This variant lies in the MYBPC3 gene (transcript NM_000256.3) at coding-DNA position 2557, where G is replaced by T; at the protein level this means replaces glycine at residue 853 with cysteine — a missense variant. Submitter rationale: Variant summary: MYBPC3 c.2557G>T (p.Gly853Cys) results in a non-conservative amino acid change located in the Fibronectin type III domain of the encoded protein sequence. Four of four in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 1.2e-05 in 245572 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance.
