This variant lies in the CRB1 gene (transcript NM_201253.3) at coding-DNA position 3901, where T is replaced by C; at the protein level this means replaces cysteine at residue 1301 with arginine — a missense variant. Submitter rationale: Variant summary: CRB1 c.3901T>C (p.Cys1301Arg) results in a non-conservative amino acid change located in the EGF-like domain (IPR000742) of the encoded protein sequence. Five of five in-silico tools predict a damaging effect of the variant on protein function. The variant was absent in 251440 control chromosomes. c.3901T>C has been reported in the literature in one homozygous individual affected with Retinal Dystrophy (Liu_2021). To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
