This variant lies in the PKHD1 gene (transcript NM_138694.4) at coding-DNA position 5336, where A is replaced by G; at the protein level this means replaces asparagine at residue 1779 with serine — a missense variant. Submitter rationale: Variant summary: PKHD1 c.5336A>G (p.Asn1779Ser) results in a conservative amino acid change in the encoded protein sequence. Five of five in-silico tools predict a benign effect of the variant on protein function. The variant was absent in 251246 control chromosomes (gnomAD). c.5336A>G has been reported in the literature as a biallelic genotype in individuals affected with Polycystic Kidney And Hepatic Disease (Melchionda_2016). These data indicate that the variant is likely to be associated with disease. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
