Uncertain significance — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_017654.4(SAMD9):c.3402G>A (p.Trp1134Ter), citing LabCorp Variant Classification Summary - May 2015: Variant summary: SAMD9 c.3402G>A (p.Trp1134X) results in a premature termination codon, predicted to cause a truncation of the encoded protein or absence of the protein in a gene where mechanism of disease is gain of function. The variant was absent in 250922 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.3402G>A in individuals affected with Normophosphatemic familial tumoral calcinosis and no experimental evidence demonstrating its impact on protein function have been reported. No clinical diagnostic laboratories have submitted clinical-significance assessments for this variant to ClinVar after 2014. Based on the evidence outlined above, the variant was classified as uncertain significance.
