NM_017950.4(CCDC40):c.2619+45G>A was classified as Pathogenic for Primary ciliary dyskinesia; Primary ciliary dyskinesia 15 by The Research Institute of Tuberculosis, Japan Anti-Tuberculosis Association, citing ACMG Guidelines, 2015: Total RNA-seq analysis of the nasal mucosal specimen with NM_017950.4:c.2619+45G>A allele revealed the activation of an aberrant splicing donor in intron 15, specifically located 50-bp downstream of the canonical splice donor site. The subsequent Sanger sequencing of the RT-PCR product confirmed the insertion of the 50-bp sequence in CCDC40 cDNA, leading to a frame shift and a premature stop codon.
