Uncertain significance for Maturity-onset diabetes of the young type 1 — the classification assigned by Foundation for Research in Genetics and Endocrinology, FRIGE's Institute of Human Genetics to NM_175914.5(HNF4A):c.128A>G (p.Asp43Gly), citing ACMG Guidelines, 2015: A heterozygous missense variant in exon 2 of the HNF4A gene that results in the amino acid substitution of Glycine for Aspartic acid at codon 65 (p.Asp65Gly) was detected. The p.Asp65Gly variant has not been reported in the 1000 genomes, gnomAD (v3.1), gnomdAD (v2) and topmed databases. The in silico predictions of the variant are probably damaging by PolyPhen-2 (HumDiv) and damaging by SIFT, LRT and MutationTaster2. The reference codon is conserved across species. In summary, the variant meets our criteria to be classified as a variant of uncertain significance.
