This variant lies in the POLH gene (transcript NM_006502.3) at 3 bases into the intron immediately after coding-DNA position 490, where A is replaced by G. Submitter rationale: Variant summary: POLH c.490+3A>G alters a non-conserved nucleotide located close to a canonical splice site and therefore could affect mRNA splicing, leading to a significantly altered protein sequence. Several computational tools predict a significant impact on normal splicing: 3 of 4 tools predict the variant weakens a 5' donor site. However, these predictions have yet to be confirmed by functional studies. The variant allele was found at a frequency of 2.4e-05 in 251324 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.490+3A>G has been reported in the literature in at least one compound heterozygous individual affected with Xeroderma Pigmentosum (e.g. Fassihi_2016).
