It has been assumed that this is a tandem duplication in direct orientation (Richardson_GIM_2018, Newman_AJHG_2015). Although exact breakpoints of this CNV are not known, it is expected to result in a large duplication change including the last exon of the C16orf35 gene. The variant was absent in 21686 control chromosomes (gnomAD, Structural Variants Dataset). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.(924+1_925-1)_(*905_?)dup in individuals affected with Epilepsy, Familial Focal, With Variable Foci 3 and no experimental evidence demonstrating its impact on protein function have been reported.
