This variant lies in the CFTR gene (transcript NM_000492.4) at coding-DNA position 490, where A is replaced by C; at the protein level this means replaces threonine at residue 164 with proline — a missense variant. Submitter rationale: Variant summary: CFTR c.490A>C (p.Thr164Pro) results in a non-conservative amino acid change in the encoded protein sequence. Five of five in-silico tools predict a damaging effect of the variant on protein function. 4/4 computational tools predict no significant impact on normal splicing. However, these predictions have yet to be confirmed by functional studies. The variant was absent in 249460 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, c.490A>C has not been reported in the literature in individuals affected with Cystic Fibrosis. and no experimental evidence demonstrating an impact on protein function has been reported.
