This variant lies in the RASA1 gene (transcript NM_002890.3) at coding-DNA position 353, where A is replaced by G; at the protein level this means replaces lysine at residue 118 with arginine — a missense variant. Submitter rationale: This RASA1 missense variant (rs1357841658) is rare (<0.1%) in a large population dataset (gnomAD v2.1.1: 1/251140 total alleles; 0.0004%; no homozygotes). It has not been reported in ClinVar nor the literature, to our knowledge. Two bioinformatic tools queried predict that this substitution would be tolerated, and the lysine residue at this position is evolutionarily conserved across many species assessed, but seven species have arganine at this position. We consider the clinical significance of c.353A>G; p.Lys118Arg in RASA1 to be uncertain at this time.
