Uncertain significance — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_032551.5(KISS1R):c.1028_1041del (p.Arg343fs), citing LabCorp Variant Classification Summary - May 2015: Variant summary: KISS1R c.1028_1041del14 (p.Arg343ProfsX95) causes a frameshift which results in an extension of the protein. At-least one protein extension have been reported in association with hypogonadotropic hypogonadism (PMID 16322390). The variant was absent in 139802 control chromosomes (gnomAD). To our knowledge, no occurrence of c.1028_1041del14 in individuals affected with Hypogonadotropic Hypogonadism 8 With Or Without Anosmia and no experimental evidence demonstrating its impact on protein function have been reported. No clinical diagnostic laboratories have submitted clinical-significance assessments for this variant to ClinVar after 2014. The available data is not sufficient to make conclusions about the variant significance. Based on the evidence outlined above, the variant was classified as uncertain significance.
