Uncertain significance — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NC_000023.10:g.(?_31137344)_(31341776_31366672)dup, citing LabCorp Variant Classification Summary - May 2015: Variant summary: The variant identified by MLPA or other technology involves the duplication of exons 62-79 in the DMD gene, and includes the last exon. A presumed nomenclature of c.(9163+1_9164-1)_(*2692_?)dup has been designated for the purposes of this classification. It has been assumed that this is a tandem duplication in direct orientation (Richardson_GIM_2018, Newman_AJHG_2015). The variant was absent in 16120 control chromosomes (gnomAD, Structural Variants dataset). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. Duplication of exons 62-79 has been reported in the literature in an unaffected female carrier and her healthy 41-year-old brother (Chin_2021). To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. Two ClinVar submitters (evaluation after 2014) cite the variant as uncertain significance (n=1) and as benign (n=1). Based on the evidence outlined above, the variant was classified as uncertain significance.
