Submitter rationale: Variant summary: NAGLU c.1493T>C (p.Leu498Pro) results in a non-conservative amino acid change located in the Alpha-N-acetylglucosaminidase, C-terminal domain (IPR024732) of the encoded protein sequence. Four of five in-silico tools predict a damaging effect of the variant on protein function. The variant was absent in 231378 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.1493T>C has been reported in the literature in at least one compound heterozygous individual affected with Mucopolysaccharidosis Type IIIB (Sanfilippo Syndrome B, Lin_2018).
