NM_005654.6(NR2F1):c.463+6T>C was classified as Uncertain significance for Bosch-Boonstra-Schaaf optic atrophy syndrome by Broad Center for Mendelian Genomics, Broad Institute of MIT and Harvard, citing ACMG Guidelines, 2015: The heterozygous c.463+6T>C variant in NR2F1 was identified by our study in one individual with abnormalities of the corpus callosum. Trio exome analysis showed this variant to be de novo. The c.463+6T>C variant in NR2F1 has not been previously reported in individuals with Bosch-Boonstra-Schaaf optic atrophy syndrome. This variant was absent from large population studies. This variant is located in the 5' splice region. Computational tools do not suggest an impact to splicing. However, this information is not predictive enough to rule out pathogenicity. In summary, the clinical significance of the c.463+6T>C variant is uncertain.
