Likely pathogenic for Tooth agenesis, selective, 7; Tooth agenesis — the classification assigned by Research Center of Stomatology, College of Stomatology, Xi'an Jiaotong University to NM_002336.3(LRP6):c.3095G>T (p.Cys1032Phe), citing ACMG Guidelines, 2015: The Cys1032Phe variant in LRP6 was identified in 1 Chinese family with autosomal dominant tooth agenesis, segregated with the disease in two family members, and was absent from large population studies. The variant targets a highly conserved YWTD motif in LRP6 protein and is predicted to be deleterious by multiple bioinformatic algorithms. Structural modeling suggest that the Cys1032Phe variant disrupts local hydrogen bonding. In summary, the Cys1032Phe variant meets the ACMG criteria to be classified as likely pathogenic based upon segregation, absence from controls, targeting functional motif and bioinformatic prediction evidence.
