NM_004415.4(DSP):c.4228_4229del (p.Lys1410fs) was classified as Likely Pathogenic for Arrhythmogenic cardiomyopathy with wooly hair and keratoderma by All of Us Research Program, National Institutes of Health, citing ACMG Guidelines, 2015: The c.4228_4229del (p.Lys1410Glufs*29) variant in the DSP gene results in a frameshift and creates a premature translation stop codon. This change is predicted to result in an absent or disrupted protein product. Loss-of-function variants in DSP are known to be pathogenic (PMID: 20716751, 24503780, 25227139). This variant is absent in the general population according to gnomAD. For these reasons, the c.4228_4229del (p.Lys1410Glufs*29) variant in the DSP gene has been classified as likely pathogenic.
