This variant lies in the PYROXD1 gene (transcript NM_024854.5) at coding-DNA position 334 through coding-DNA position 337, deleting 4 bases; at the protein level this means shifts the reading frame starting at leucine residue 112, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: For these reasons, this variant has been classified as Pathogenic. This premature translational stop signal has been observed in individual(s) with clinical features of myofibrillar myopathy (PMID: 33694278). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. This variant is present in population databases (rs765752811, gnomAD 0.004%). This sequence change creates a premature translational stop signal (p.Leu112Valfs*8) in the PYROXD1 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in PYROXD1 are known to be pathogenic (PMID: 27745833).
