Submitter rationale: This variant causes a G>A nucleotide substitution at the +1 position of intron 3 of the PCSK9 gene. Splice site prediction tools predict that this variant may have a significant impact on RNA splicing, which may result in the loss of PCSK9 function and have a protective effect against familial hypercholesterolemia. To our knowledge, functional studies have not been reported for this variant. This variant has not been reported in individuals affected with PCSK9-related disorders in the literature.
