Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_001211.6(BUB1B):c.1384G>C (p.Glu462Gln), citing Ambry Variant Classification Scheme 2023: The p.E462Q variant (also known as c.1384G>C), located in coding exon 10 of the BUB1B gene, results from a G to C substitution at nucleotide position 1384. The glutamic acid at codon 462 is replaced by glutamine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
