Uncertain significance — the classification assigned by Ambry Genetics to NM_006904.7(PRKDC):c.3170A>C (p.Lys1057Thr), citing Ambry Variant Classification Scheme 2023: The p.K1057T variant (also known as c.3170A>C), located in coding exon 27 of the PRKDC gene, results from an A to C substitution at nucleotide position 3170. The lysine at codon 1057 is replaced by threonine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
