Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_001378454.1(ALMS1):c.9017A>T (p.His3006Leu), citing Ambry Variant Classification Scheme 2023: The p.H3007L variant (also known as c.9020A>T), located in coding exon 10 of the ALMS1 gene, results from an A to T substitution at nucleotide position 9020. The histidine at codon 3007 is replaced by leucine, an amino acid with similar properties. This amino acid position is poorly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
