NM_080732.4(EGLN2):c.421A>G (p.Asn141Asp) was classified as Uncertain significance by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.N141D variant (also known as c.421A>G), located in coding exon 1 of the EGLN2 gene, results from an A to G substitution at nucleotide position 421. The asparagine at codon 141 is replaced by aspartic acid, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
