Uncertain significance — the classification assigned by Ambry Genetics to NM_006393.3(NEBL):c.272G>A (p.Gly91Asp), citing Ambry Variant Classification Scheme 2023: The p.G91D variant (also known as c.272G>A), located in coding exon 4 of the NEBL gene, results from a G to A substitution at nucleotide position 272. The glycine at codon 91 is replaced by aspartic acid, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
