Uncertain significance — the classification assigned by Ambry Genetics to NM_001365951.3(KIF1B):c.696T>A (p.Asn232Lys), citing Ambry Variant Classification Scheme 2023: The p.N232K variant (also known as c.696T>A), located in coding exon 6 of the KIF1B gene, results from a T to A substitution at nucleotide position 696. The asparagine at codon 232 is replaced by lysine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
