Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_017849.4(TMEM127):c.143T>A (p.Leu48His), citing Ambry Variant Classification Scheme 2023: The p.L48H variant (also known as c.143T>A), located in coding exon 1 of the TMEM127 gene, results from a T to A substitution at nucleotide position 143. The leucine at codon 48 is replaced by histidine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
