Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_006231.4(POLE):c.5457G>A (p.Met1819Ile), citing Ambry Variant Classification Scheme 2023: The p.M1819I variant (also known as c.5457G>A), located in coding exon 40 of the POLE gene, results from a G to A substitution at nucleotide position 5457. The methionine at codon 1819 is replaced by isoleucine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
