Uncertain significance — the classification assigned by Ambry Genetics to NM_002519.3(NPAT):c.95C>T (p.Ser32Leu), citing Ambry Variant Classification Scheme 2023: The p.S32L variant (also known as c.95C>T), located in coding exon 2 of the NPAT gene, results from a C to T substitution at nucleotide position 95. The serine at codon 32 is replaced by leucine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
