Uncertain significance — the classification assigned by Ambry Genetics to NM_006393.3(NEBL):c.1099C>G (p.Gln367Glu), citing Ambry Variant Classification Scheme 2023: The p.Q367E variant (also known as c.1099C>G), located in coding exon 11 of the NEBL gene, results from a C to G substitution at nucleotide position 1099. The glutamine at codon 367 is replaced by glutamic acid, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
