Uncertain significance for Dilated cardiomyopathy 1R; Hypertrophic cardiomyopathy 11; Atrial septal defect 5 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_005159.5(ACTC1):c.991A>T (p.Ile331Phe), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces isoleucine, which is neutral and non-polar, with phenylalanine, which is neutral and non-polar, at codon 331 of the ACTC1 protein (p.Ile331Phe). This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with ACTC1-related conditions. ClinVar contains an entry for this variant (Variation ID: 2486898). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be disruptive. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
