Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_003900.5(SQSTM1):c.620G>A (p.Ser207Asn), citing Ambry Variant Classification Scheme 2023: The c.620G>A (p.S207N) alteration is located in exon 4 (coding exon 4) of the SQSTM1 gene. This alteration results from a G to A substitution at nucleotide position 620, causing the serine (S) at amino acid position 207 to be replaced by an asparagine (N). This allele was reported in one heterozygous individual in population-based cohorts in the Genome Aggregation Database (gnomAD). This amino acid position is highly conserved in available vertebrate species. This alteration is predicted to be tolerated by in silico analysis. Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear.
