Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_002485.5(NBN):c.2234+2T>C, citing Ambry Variant Classification Scheme 2023. This variant lies in the NBN gene (transcript NM_002485.5) at the canonical splice donor site of the intron immediately after coding-DNA position 2234, where T is replaced by C; at the protein level this means a change at this position may disrupt normal splicing. Submitter rationale: The c.2234+2T>C intronic variant results from a T to C substitution two nucleotides after coding exon 15 in the NBN gene. This nucleotide position is highly conserved in available vertebrate species. This alteration occurs at the 3' terminus of the NBN gene, is not expected to trigger nonsense-mediated mRNA decay, and only impacts the last 25 amino acids of the protein. The exact functional effect of this alteration is unknown. In silico splice site analysis predicts that this alteration will weaken the native splice donor site; however, direct evidence is insufficient at this time (Ambry internal data).
