NM_003265.3(TLR3):c.493G>A (p.Gly165Arg) was classified as Uncertain significance for Herpes simplex encephalitis, susceptibility to, 1 by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be disruptive. ClinVar contains an entry for this variant (Variation ID: 2479446). This variant has not been reported in the literature in individuals affected with TLR3-related conditions. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. This sequence change replaces glycine, which is neutral and non-polar, with arginine, which is basic and polar, at codon 165 of the TLR3 protein (p.Gly165Arg). This variant is not present in population databases (gnomAD no frequency).
