Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_004304.5(ALK):c.747G>A (p.Met249Ile), citing Ambry Variant Classification Scheme 2023: The p.M249I variant (also known as c.747G>A), located in coding exon 2 of the ALK gene, results from a G to A substitution at nucleotide position 747. The methionine at codon 249 is replaced by isoleucine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
