Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_001689.5(ATP5MC3):c.161A>G (p.Gln54Arg), citing Ambry Variant Classification Scheme 2023: The c.161A>G (p.Q54R) alteration is located in exon 3 (coding exon 3) of the ATP5G3 gene. This alteration results from an A to G substitution at nucleotide position 161, causing the glutamine (Q) at amino acid position 54 to be replaced by an arginine (R). This allele was reported in one heterozygous individual in population-based cohorts in the Genome Aggregation Database (gnomAD). This amino acid position is well conserved in available vertebrate species. This alteration is predicted to be tolerated by in silico analysis. Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear.
