NM_000371.4(TTR):c.337G>A (p.Val113Met) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.V113M variant (also known as c.337G>A) is located in coding exon 4 of the TTR gene. The valine at codon 113 is replaced by methionine, an amino acid with highly similar properties. This change occurs in the first base pair of coding exon 4. This variant was reported in an adult individual with sporadic pure motor and bulbar neuropathy with amyloid deposits on nerve biopsy (Lozeron P et al. Amyloid, 2013 Sep;20:188-92). Note, this variant is also referred to as V93M in the literature. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
