NM_004415.4(DSP):c.3004C>T (p.Arg1002Trp) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.R1002W variant (also known as c.3004C>T), located in coding exon 22 of the DSP gene, results from a C to T substitution at nucleotide position 3004. The arginine at codon 1002 is replaced by tryptophan, an amino acid with dissimilar properties. This variant was detected in a cardiomyopathy genetic testing cohort; however, clinical details were limited, and additional cardiac variants were detected in some cases. (van Lint FHM et al. Neth Heart J, 2019 Jun;27:304-309). This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
