Uncertain significance — the classification assigned by Ambry Genetics to NM_004336.5(BUB1):c.1733A>T (p.Asp578Val), citing Ambry Variant Classification Scheme 2023: The p.D578V variant (also known as c.1733A>T), located in coding exon 16 of the BUB1 gene, results from an A to T substitution at nucleotide position 1733. The aspartic acid at codon 578 is replaced by valine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
