Uncertain significance — the classification assigned by Ambry Genetics to NM_004336.5(BUB1):c.2965T>C (p.Phe989Leu), citing Ambry Variant Classification Scheme 2023: The p.F989L variant (also known as c.2965T>C), located in coding exon 24 of the BUB1 gene, results from a T to C substitution at nucleotide position 2965. The phenylalanine at codon 989 is replaced by leucine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
