NM_000051.4(ATM):c.1438T>G (p.Leu480Val) was classified as Uncertain significance by GeneDx, citing GeneDx Variant Classification (06012015). This variant lies in the ATM gene (transcript NM_000051.4) at coding-DNA position 1438, where T is replaced by G; at the protein level this means replaces leucine at residue 480 with valine — a missense variant. Submitter rationale: This variant is denoted ATM c.1438T>G at the cDNA level, p.Leu480Val (L480V) at the protein level, and results in the change of a Leucine to a Valine (TTA>GTA). This variant has not, to our knowledge, been published in the literature as pathogenic or benign. ATM Leu480Val was not observed in approximately 6,500 individuals of European and African American ancestry in the NHLBI Exome Sequencing Project, suggesting it is not a common benign variant in these populations. Since Leucine and Valine share similar properties, this is considered a conservative amino acid substitution. ATM Leu480Val occurs at a position that is conserved across species and is not located in a known functional domain (Tavtigian 2009, Stracker 2013). In silico analyses predict that this variant is probably damaging to protein structure and function. Based on currently available evidence, it is unclear whether ATM Leu480Val is a pathogenic or benign variant. We consider it to be a variant of uncertain significance.
