This variant lies in the ATM gene (transcript NM_000051.4) at coding-DNA position 9088, where G is replaced by A; at the protein level this means replaces glycine at residue 3030 with arginine — a missense variant. Submitter rationale: This variant is denoted ATM c.9088G>A at the cDNA level, p.Gly3030Arg (G3030R) at the protein level, and results in the change of a Glycine to an Arginine (GGA>AGA). This variant has not, to our knowledge, been published in the literature as pathogenic or benign. ATM Gly3030Arg was not observed in approximately 6,500 individuals of European and African American ancestry in the NHLBI Exome Sequencing Project, suggesting it is not a common benign variant in these populations. Since Glycine and Arginine differ in polarity, charge, size or other properties, this is considered a non-conservative amino acid substitution. ATM Gly3030Arg occurs at a position that is conserved across species and is located in the FATC domain (Stracker 2013). In silico analyses predict that this variant is probably damaging to protein structure and function. Based on currently available evidence, it is unclear whether ATM Gly3030Arg is a pathogenic or benign variant. We consider it to be a variant of uncertain significance.
