NM_030962.4(SBF2):c.304G>T (p.Gly102Cys) was classified as Uncertain significance by GeneDx, citing GeneDx Variant Classification (06012015). This variant lies in the SBF2 gene (transcript NM_030962.4) at coding-DNA position 304, where G is replaced by T; at the protein level this means replaces glycine at residue 102 with cysteine — a missense variant. Submitter rationale: The G102C variant has not been published as a pathogenic variant, nor has it been reported as a benign polymorphism to our knowledge. It was not observed in approximately 6,500 individuals of European and African American ancestry in the NHLBI Exome Sequencing Project, indicating it is not a common benign variant in these populations. The G102C variant is a non-conservative amino acid substitution, which is likely to impact secondary protein structure as these residues differ in polarity, charge, size and/or other properties. In silico analysis predicts this variant is probably damaging to the protein structure/function. However, this substitution occurs at a position that is not conserved across species and missense variants in nearby residues have not been reported in the Human Gene Mutation Database (Stenson et al., 2014). Therefore, based on the currently available information, it is unclear whether this variant is a pathogenic or a rare benign variant.
